Cagrilintide (AM833, NN9838) is an investigational long-acting amylin receptor agonist developed by Novo Nordisk and studied for chronic weight management, both as a monotherapy and co-administered with semaglutide (the combination known as CagriSema). It is NOT FDA-approved for any indication as of June 2026. It is built on a modified pramlintide-like backbone with an attached fatty diacid that extends its elimination half-life to roughly 159 to 195 hours (around 180 hours, about 7 days), which supports once-weekly subcutaneous dosing. Because a human half-life in this range is reported, this calculator can model weekly plasma accumulation, with steady state reached over roughly 5 to 6 weeks of weekly dosing. The 2.4 mg figure used as the default reflects the highest fixed dose paired with semaglutide in trials and the second-highest monotherapy dose; it is a reference figure from clinical studies, has no clinically established basis for general use, and is not a recommendation. This calculator performs math on the values you enter and does not recommend any dose.

Common questions

How do I reconstitute cagrilintide?

Add your chosen volume of bacteriostatic water to the lyophilized vial, directing the stream gently against the glass wall, and swirl slowly (do not shake) until the powder is fully dissolved. The calculator above converts your vial size and water volume into the exact draw volume and units for your target weekly dose. Store the reconstituted solution refrigerated and use within the period recommended by your supplier.

What is the cagrilintide dosage?

Cagrilintide is investigational and has no FDA-approved dosing guideline. In published phase 2 work, once-weekly subcutaneous doses ranged from 0.3 mg to 4.5 mg, and the CagriSema combination has paired 2.4 mg cagrilintide with 2.4 mg semaglutide. These figures come from clinical trials, not approved prescribing information, and are not a recommendation. This calculator performs math on the values you enter; it does not recommend any dose. Consult a licensed clinician before using any investigational compound.

How many units is 2.4 mg of cagrilintide?

It depends on your reconstitution concentration. With a 10 mg vial in 2 mL of bacteriostatic water (5 mg/mL), a 2.4 mg dose is 0.48 mL, which equals 48 units on a U-100 insulin syringe. Enter your own vial size and water volume in the calculator above for your exact number.

Is cagrilintide FDA-approved?

No. Cagrilintide (AM833) is not approved by the FDA for any indication. It is an investigational amylin receptor agonist studied by Novo Nordisk for weight management, both alone and combined with semaglutide as CagriSema.
